Does the remote need line of sight to the Roku box?

If your Roku is on your network you can add it as a Wi-Fi device and then they can be hidden and not use IR (with line of sight)

Could this work with the xiaomi mii box s remote?

I don't know it works fine with the alexa voice fire tv remote

Will this remote control the power on and off on my Samsung tv and my onkyo receiver and the volume up and down and mute on receiver??

yep! that's precisely my setup (samsung tv and yamaha receiver). One power button controls tv on/off, the other power button controls receiver on/off, and the volume and mute buttons control receiver sound. works perfectly. :)
